Heat shock protein 90 of Bonamia ostreae: characterization and possible correlation with infection of the flat oyster, Ostrea edulis ArchiMer

In this study we described the cytosolic HSP90 of Bonamia ostreae, an intracellular parasite of Ostrea edulis haemocytes. The complete open reading frame was assembled by Rapid Amplification cDNA Ends reactions on cDNA of B. ostreae infected haemocytes. HSP90 amplification was corroborated in infected oysters and B. ostreae purified cells. The functionality of the HSP90, studied by inhibitory assays with radicicol, suggests that this protein may play a role in haemocyte invasion. Our results inform the molecular basis that governs B. ostreae-O. edulis interactions.

Bonamiosis, favoured by change of temperature and stress, causes mortalities in the adult flat oyster Ostrea edulis in the majority of European oyster farmers. The life cycle of this Haplosporidia protozoan Bonamia ostreae is unknown, it is not dttectable during the five mon th latent period. Because of the Jack of sensibility of reference techniques, histology and tissue smear, further use of PCR assays or HCM-CC, in particular, is recommended. Analysis of data collected in France from 1989 to 2004 shows the absence of simple prediction methods and an apparent cyclicity of the disease. Les parasites du groupe "Mikrocell" : Qui est qui? ArchiMer

Bonamiose, a disease caused by the protozoa Bonamia ostreae, has destroyed the flat oyster Ostrea edulis production, in France but also in Europe and in the United States since 1979. Another species of Bonamia has been monitored in New-Zealand, in Australia and lately in Chile. In Canada and Australia, two other parasites have been described in cupped oyster: Mikrocytos mackini and Mikrocytos roughleyi. All these small size protozoa are grouped under the name "Mikrocell". For most of them, light and electronic microscope descriptions have been realised. Based on those criteria, different taxonomic positions have been proposed. However, these analysis are still questioned today.

Pathology of cultured paua Haliotis iris infected with a novel haplosporidian parasite, with some observations on the course of disease ArchiMer

Mortalities among juvenile paua Haliotis iris Martyn 1784 in a commercial culture facility were reported in April 2000. Histology of moribund paua showed heavy systemic infections of a uni- to multi-nucleate stage of a novel organism later confirmed by transmission electron microscopy (TEM) and molecular studies to be a haplosporidian. Multinucleate plasmodia up to 25 μm diameter with up to 17 nuclei were detectable in wet preparations of hemolymph from heavily infected paua. The presence of the haplosporidian in the affected facility was associated with mortalities of slow growing 'runt' paua during the summer months. Registro de Nephridiophaga sp. (Protista: Nephridiophagidae) en Apis mellifera (Hymenoptera: Apidae) del Sur de la región Pampeana Rev. Soc. Entomol. Argent.

Durante estudios prospectivos tendientes a la detección de protistas asociados a ápidos en la región Pampeana, se observó la presencia de esporos ovales bicóncavos y grupos de esporos (cúmulos) en los túbulos de Malpighi de abejas de Dufaur, partido de Saavedra, sudoeste de la provincia de Buenos Aires. Los esporos maduros midieron 4,8 ± 0,05 x 2,4 ± 0,03 μm y la carga (intensidad) promedió 5,71 ± 1,49 x 10(6) esporos/abeja. Las detecciones se efectuaron entre julio y octubre de 2006 y la prevalencia en las colmenas positivas osciló entre 1 y 16,7 %. Las características morfológicas de los esporos, el lugar de desarrollo y la especie huésped involucrada sugieren que el microorganismo en cuestión, pertenece al género Nephridiophaga y sería N. apis...

Whole-genome amplification: a useful approach to characterize new genes in unculturable protozoan parasites such as Bonamia exitiosa ArchiMer

Bonamia exitiosa is an intracellular parasite (Haplosporidia) that has been associated with mass mortalities in oyster populations in the Southern hemisphere. This parasite was recently detected in the Northern hemisphere including Europe. Some representatives of the Bonamia genus have not been well categorized yet due to the lack of genomic information. In the present work, we have applied Whole-Genome Amplification (WGA) technique in order to characterize the actin gene in the unculturable protozoan B. exitiosa. This is the first protein coding gene described in this species. Molecular analysis revealed that B. exitiosa actin is more similar to Bonamia ostreae actin gene-1.
